I want to make a scupper access plate on my rear transit wall behind my transom door .on my 96 272. Any body do this before

"Transit"? Can you post a picture of what you're talking about? Or give a better description?
But if you're simply talking about drilling a hole in a relatively non structural area... that's all it really is... Drill/cut your hole and install the plate. It's pretty basic.

The addition of an access plate is fairly common when a scupper hose or fitting needs replacement and it isn't east to get to. Just adding one for possible future need is not common at all.
Ah, "transom"... well, technically I guess it's still the hull, but close enough. Yes, that area would be fine. Mark your hole, cut, install plate, go fishing. I'm just guestimating since I'm not there to actually measure, but there looks like there is plenty of room for even an 8" plate. Stay away from edges and corners, though, by at least a few inches. Center it and you're golden.
I think he needs a water tight access in that location. Take a large saucer, turn it upside down, and make sure the area is flat where the hole goes.

See if you can find a "cutout" size or a "template" for it. Cut a piece of paper or cardboard to that size and tape it onto the proposed location and see how much "meat" is left of the hull.
Quick tip... when you cut a corner, don't make it a hard corner - always use a drill bit to make a rounded corner (say 1/2". This keeps more strength in the panel and also helps to prevent spider cracking of the gelcoat (it's cosmetic, but still).

I’m going to use most likely an 8 inch circle and I have a hand router I’m going to make a jig out of the cover plate then screw it down and trace the middle with the router

I’m still no sure about what I want to put there but I do need access to scuppers. Just throwing ideas around!!!
Someone showed cutting access ports on their swim platform, on one of the scupper threads.

Pictures of what? More detail. Are you looking for pictures of someone cutting a hole? I've cut holes in boats more times than I can remember, but I never thought to take pics because it's so straight forward - not something you really need a pic for, you know?
Would it better to cut a hole just inside/below the transom door?
I was asking Anybody cut a hole there before that’s what I want to picture of to get an idea but basically I know what to do . I was also thinking below The transom door also!!

That seat in your cockpit... is whatever is inside that seat removable? Just wondering if that would give you access to the ones on the floor.
